为了改善蛋白质的功能特性,该研究分析了超声复合酸处理对花生分离蛋白的溶解性、紫外光谱、荧光光谱、二级结构及纳米结构等的影响,并探讨不同处理条件下蛋白聚集体结构变化的机理。结果表明,超声作用可以明显促进花生分离蛋白的不溶性聚集体向可溶性聚集体转变,单独超声及超声复合酸处理使其溶解度相对于对照分别增加了12.9%和15.3%(P<0.05);电泳、紫外和荧光光谱表明,超声和酸作用均有助于亚基解离及蛋白质结构展开,从而促进更多的酪氨酸、色氨酸和苯丙氨酸等疏水性基团暴露;圆二色谱分析显示,与对照相比,在超声复合酸处理使花生分离蛋白的a-螺旋增加21.9%,β-折叠减少3.6%,无规则卷曲增加1.8%(P<0.05);纳米结构表明,超声复合酸处理最大程度地降低了花生分离蛋白的颗粒大小。该研究证实,在酸性条件下进行超声处理,能显著促进花生分离蛋白的亚基解离和结构展开。该研究为后期蛋白亚基重新相互作用形成不同功能的改性蛋白提供参考。  相似文献

Under rainfed agriculture without supplementary supply, crop failure due to erratic rainfall has become a common phenomenon in many regions of Ethiopia. Spate irrigation development with storage provision at the 5,000 ha Boru Dodota Spate Irrigation Scheme is one of the initiatives to move away from exclusively rain-dependent agriculture. This initiative has faced several challenges. Lack of design experience and failure to fully grasp the unpredictable nature of the spate flow caused the Boru Dodota Spate Irrigation Scheme to be implemented without considering the necessity of storage ponds. In addition, absence of a systematic approach to assist planners has resulted in non-optimal design of ponds’ capacity and operation. The study, on which this article is based, aimed at optimum storage operation to irrigate the maximum possible area with the existing ponds’ capacity and available water resource. During the study, the surface storage and irrigation scheme planning model was developed and used to analyze several pond operation scenarios in Boru Dodota Spate Irrigation Scheme. The main findings were as follows: (1) Supplementing the rainfall with the operation of the existing 19 ponds that enable weekly irrigation frequency results in irrigating at least 6,600 ha. (2) An increase in the number of ponds does not always guarantee an increase in the size of irrigated land because the water resources, the operation, and management are defining factors. (3) It is not economical to only rely on spate flow for irrigation as even with 200 ponds, a maximum of 1,250 ha could be irrigated.  相似文献

This interlaboratory study evaluated a real-time multiplex polymerase chain reaction (PCR) method for identification of salmon and trout species in a range of commercial products in North America. Eighty salmon and trout products were tested with this method by three independent laboratories. Samples were collected in the United States and Canada, and only the collecting institution was aware of the species declaration. Following analysis with real-time PCR, all three laboratories were able to identify species in 79 of the 80 products, with 100% agreement on species assignment. A low level of fraud was detected, with only four products (5%) found to be substituted or mixtures of two species. The results for two of the fraudulent products were confirmed with alternate methods, but the other two products were heavily processed and could not be verified with methods other than real-time PCR. Overall, the results of this study show the usefulness and versatility of this real-time PCR method for the identification of commercial salmon and trout species.  相似文献

A set of 77 markers was used to describe the genetic diversity in a group of 58 tetraploid wheat accessions. Analysis was performed using 31 neutral SSR markers, 31 SSR/STS markers linked with reported major stem rust resistance genes and 15 SSR markers linked with QTL identified for resistance to Ethiopian stem rust races of Puccinia graminis Pers. f. sp. tritici Eriks. et E. Henn. (Pgt),including Ug99. The material consisted of 32 (Triticum durum s.l. incl. T. aethiopicum Jakubz., Triticum turgidum and Triticum polonicum) landraces and 22 registered T. durum varieties from Ethiopia that were released 1966–2009 and four T. durum varieties from ICARDA. A total of 720 alleles were detected. Considering the three marker sets, the mean number of alleles was higher for major stem rust resistance gene linked markers (9.9) followed by neutral SSR markers (9.2) and markers linked with QTL for stem rust resistance (8.5). Dendrograms derived from UPGMA analysis grouped the accessions into two major clusters. The principal component analysis based on the combination of the three marker sets formed three groups. The 1st group was composed of all the improved varieties, whereas the 2nd and the 3rd group contained the landraces. All the landraces that formed the 3rd group were susceptible to Ethiopian stem rust races of Pgt including Ug99. The information on the extent of the genetic diversity of the improved varieties obtained in this investigation will be helpful for developing appropriate breeding strategies to broadening the genetic base of durum wheat varieties in further breeding programmes.  相似文献

为了确定监测时长、监测网具、站位布局对禁捕后长江鱼类资源监测评估的有效性,于2021年5—7月在长江中游10个站位开展了鱼类资源捕捞监测,每个站位连续监测15 d,从日捕获量、物种记录数、渔获物群落结构等3方面着手,对监测时长、监测网具、站位布局等的设置进行探讨。结果显示,连续11 d捕捞监测所得累计日均渔获量基本达到稳定,连续15 d捕捞监测可以记录到站位近70%的鱼类种类数,所得累计鱼类群落结构基本达到稳定。网具类型、规格的使用覆盖对监测结果中鱼类种类记录数、鱼类群落结构有明显影响,网具使用量对监测结果中日均渔获量有明显影响。10个监测站位间的鱼类群落结构具有明显差异,结合监测站位间的空间距离来看,鱼类群落结构具有较高的空间异质性。研究表明,为了保障评估结果的准确性,基于原始监测数据进行相关评估之前,有必要对监测数据的充分性、有效性进行检验,网具类型、规格和使用量对监测结果的影响应予以适当考虑,各具体江段的鱼类群落结构评估应由相应具体江段的监测结果来支撑。本研究将为长江流域重点水域全面禁捕后的水生生物资源监测评估提供科学支撑。  相似文献

Soil degradation in the highlands of Ethiopia is a serious problem that threatens the sustainability of agriculture. Although efforts have been made to develop and promote several soil conservation technologies, their adoption has not been widespread. This study, based on responses from a sample of 101 households which operate on 204 plots in Western Ethiopia, interviewed in 2003 cropping year, employed a Tobit model to identify the main factors influencing farmers' decision to adopt and intensify the use of improved and indigenous soil and water conservation (SWC) measures. The results suggest that area of cultivated land, land/labour ratio, age and education level of household head and distance of the plot from home and slope of the plot provided statistically adequate predictive power on the use and intensity of SWC measures. Global maps of thermal and fast neutron fluxes from the moon suggest three end-member compositional units. A high thermal and low fast neutron flux unit correlates with the lunar highlands and is consistent with feldspathic rocks. The South Pole-Aitken basin and a strip that surrounds the nearside maria have intermediate thermal and fast neutron flux levels, consistent with more mafic rocks. There appears to be a smooth transition between the most mafic and feldspathic compositions, which correspond to low and high surface altitudes, respectively. The maria show low thermal and high fast neutron fluxes, consistent with basaltic rocks.  相似文献

The Lunar Prospector neutron spectrometer data correlate well with iron and titanium abundances obtained through analysis of Clementine spectral reflectance data. With the iron and titanium dependence removed, the neutron spectrometer data also reveal regions with enhanced amounts of gadolinium and samarium, incompatible rare earth elements that are enriched in the final phases of magma crystallization. These regions are found mainly around the ramparts of the Imbrium impact basin but not around the other basins, including the much larger and deeper South Pole-Aitken basin. This result confirms the compositional uniqueness of the surface and interior of the Imbrium region.  相似文献
